1. Phylogenetic Position

mtDNA haplogroups are defined by maternal inheritance.

D4 is one of the most common East Asian mitochondrial lineages. D4e is a sub-branch defined by additional specific mtDNA mutations.

2. Geographic Distribution

D4e is predominantly found in:

  • Korea
  • Japan
  • Northern and Eastern China
  • Parts of Mongolia and Siberia
It is considered an East Asian maternal lineage, likely expanding during Upper Paleolithic or early Hololithic population movements in Northeast Asia.  

4. Biological Significance

Mitochondria:

  • Contain their own circular genome (~16,569 bp)
  • Encode 13 oxidative phosphorylation proteins
  • Are inherited strictly maternally
D4e itself is not a disease classification. It is a population-genetic lineage marker.

Some D4 subtypes have been studied in relation to:

  • Longevity
  • Metabolic efficiency
  • Adaptation to cold climates
  • Neurodegenerative disease susceptibility
However, D4e specifically does not have a well-established unique pathological phenotype.

5. Functional Considerations

mtDNA haplogroups can influence:

  • ROS production
  • Mitochondrial membrane potential
  • ATP generation efficiency
  • Apoptotic sensitivity
But these effects are typically subtle and population-level rather than deterministic at the individual level.
